Isolation and inhibitory activity of gabaculine, a new potent inhibitor of gamma-aminobutyrate aminotransferase produced by a Streptomyces.

چکیده
It is well known that GABA plays an inhibitory role in crustacean ans mammalian central nervous system [ 1,2]. Since GABA does not cross the blood-brain barrier, various inhibitors of GABA aminotransferase which cause an increase in its cerebral level have been introduced into this field of research [3-121. Most of these inhibitors are very toxic and not specific for GABA aminotransferase. During our search for new inhibitors of GABA aminotransferase from microbial origin, a potent inhibitor (designated as gabaculine) was found to be present in the culture of a Streptomyces. The present paper describes isolation and inhibitory activity of gabaculine. The results indicate that this new microbial metabolite is a potent, competitive inhibitor of GABA aminotransferase from both beef brain and Pseudomonas ovalis, having Ki values of 0.37-0.80 PM. The physicochemical properties and identification of chemical structure of gabaculine have been reported elsewhere [13].

L-Ornithine:2-oxoacid aminotransferase is a specific enzyme with respect to the amino group donor. Nevertheless it was found that this enzyme is inhibited by some 4-aminobutyrate analogs, 4-aminohex-5-ynoic acid and 5-amino-l,%cyclohexadienyl-carboxylic acid (gabaculine), which are currently considered to be enzyme-activated irreversible inhibitors of I-aminobutyrate:2-oxoglutarate aminotransfe...

The central role of glutamate as the progenitor of A-aminolaevulinic acid for tetrapyrrole formation in oxygenic photosynthetic cells [l-41 is now well-established. The initial reaction in the conversion is the activation of glutamate by ligation to tRNA9lu. The ester-linked glutamate moiety is then reduced to glutamate-1-semialdehyde which in turn undergoes structural rearrangement to give n-a...

The application of new probiotics is a good strategy in the biological control of infectious diseases in aquaculture. Approximately 100 marine actinobacteria isolates were obtained from 10 sediment samples of shrimp farms. Heat treatment of sediment samples resulted in a selective reduction of the non actinobacterial heterotrophic microflora. Starch nitrate agar medium exhibited more efficacy t...
